The tumor microenvironment (TME) is a dynamic space that is constantly adjusting to tumor growth including numerous processes that promote tumorigenesis. The interactions involve many different cell types found in the TME including tumor cells, fibroblasts, dendritic cells, TILs, macrophages and NK cells as well as many noncellular components including the ECM and signaling molecules. These components and the spatial distribution of tumor cells, immune cells and ECM are unique to individual patients and essential to tumorigenesis. Therefore, a full understanding of the effect of a drug on tumor growth depends on understanding the interplay of all the components of the TME.
